Plant-derived exosomes contain signaling substances secreted by plant cells. Exosomes purified from plants are known to have no toxicity compared to mammalian-derived exosomes. Due to their numerous advantages and activities, they can be utilized in pharmaceuticals, cosmetics, and food.The ultracentrifugation method of isolating exosomes requires highly expensive equipment and long separation times. Tangential flow filtration is complicated in terms of equipment setup. CICA EXOSOME secures stability of plant-derived exosomes.Centella asiatica has long been used as a medicinal plant, active components of which include pentacyclic triterpene: asiaticoside, madecassoside, asiatic acid, and madecassic acid. These components are known for their efficacy in promoting collagen production in skin cells, antioxidant activity, antiaging properties, anti-inflammatory effects, photoprotective effects, and wound healing capabilities, and are used for wound healing. In this study, exosomes were extracted from Centella asiatica (CICA) and evaluated for their potential as cosmetic raw materials. Confirmation of anti-inflammatory effect of C. asiatica ExosomeThe transcription factor nuclear factor-%u03baB (NF-%u03baB) is the central mediator of pro-inflammatory gene induction. In this experiment, human epidermal keratinocyte (HEKa) cells were exposed to UVB (20 mJ/cm%u00b2) to activate NF-%u03baB expression. Cells were treated with varying concentrations of Cica extract or Cica exosome, or 0.01% Allantoin to compare NF-%u03baB expression levels. The result showed that the cells treated with Cica exosome had downregulated these two gene expressions compared to the control, indicating its higher efficacy in antiinflammatory effect compared to Cica extract. Treatment with 1x107 particles/ml of Cica exosome also showed higher efficacy than Allantoin, which is proven to suppress inflammatory responses by inhibiting the overactivation of the NF-%u03baB signaling pathways (Figure 1).Makale / Article41
